The method involving RT-PCR (reverse transcriptase polymerase chain reaction) exploits our observation that the silent copy transcripts extend beyond the regions of homology to the flanking sequences specific for the donor cassettes, thus allowing design of oligos that are specific for the different donors. The results are validated using a known silencing mutant swi6. date: 2008-01 date_type: published publication: BioTechniques volume: 44 number: 1 publisher: Informa BioSciences pagerange: 54, 56, 58 refereed: TRUE issn: 0736-6205 official_url: http://www.biotechniques.com/multimedia/archive/00003/BTN_A_000112671_O_3569a.pdf related_url_url: http://www.biotechniques.com/multimedia/archive/00003/BTN_A_000112671_O_3569a.pdf related_url_type: pub citation: Ahmed, Shakil and Singh, Jagmohan (2008) RT-PCR method for selective detection of silent gene transcripts in silencing mutants in homothallic strains of Schizosaccharomyces pombe.
