"Enzymes for the degradation of 3,5-xylenol of\r\nPseudomonas putida NCIB9869 are encoded on a transmissible\r\nplasmid, pRA500. Genes specifying the inducible\r\nsynthesis of some enzymes encoded on pRA500,\r\nnamely, citraconase (G), citramalate coenzyme-Atransferase\r\n(H), citramalyl-coenzyme-A lyase (I), maleylpyruvate\r\nisomerase (J) and fumarylpyruvate hydrolase\r\n(K) have been cloned on a 7.9-kb HindIII fragment into\r\nthe vector pKT231 to give pRA507. Biochemical and\r\nrestriction analysis of pRA507 and some of its deleted\r\nderivatives has enabled preliminary locations to be\r\nassigned to the genes encoding these enzymes. The\r\nstructural genes for enzymes H and I, together with\r\na regulatory gene controlling their expression, are\r\nlocated on a 1.6-kb HindIII/XhoI fragment of pRA507\r\nand the genes for enzymes G, J and K are located\r\nwithin a 4.5-kb XhoI/C1aI fragment of pRA507, which\r\nlies immediately adjacent to the former fragment. Biochemical\r\nanalysis of the strain carrying pRA507, of its\r\nplasmid-free derivative and of other plasmid-free derivatives\r\nof the wild-type P. putida has provided indirect\r\nevidence for the presence of two isofunctional enzymes\r\nat three diĀ¤erent steps of the degradative pathway. One\r\nset of 3-hydroxybenzoate 6-hydroxylase, gentisate 1,2-\r\ndioxygenase and maleylpyruvate hydrolase is encoded\r\non pRA500 and these enzymes are inducible only.\r\nA second set is encoded on the chromosome and these\r\nenzymes are constitutive and not further inducible. The\r\nchromosomally located genes appeared to be silent\r\nuntil 3,5-xylenol-negative strains were incubated for\r\nprolonged periods with compounds such as m-hydroxybenzoate\r\nor p-cresol."^^ .
