Abstract
A cream-white bacterial colony designated SH1-B was isolated on Tryptone Soya Agar (TSA) plated with a brackish water sample collected from the Pichavaram mangrove forest. The strain SH1-B was Gram-stain negative, rod-shaped, motile, non-spore-forming aerobic bacterium. Catalase and oxidase activities were positive. Cells grew between 15 and 37 degrees C and pH 5.0-9.0. The nucleotide sequence of 16 S rRNA gene displayed high similarity with species of the genus Pseudomonas, including P. khazarica TBZ2(T) (98.5%), P. yangonensis MY50(T) (98.0%) and P. oleovorans subsp. oleovorans DSM1045(T) (98.0%). Genome sequence analysis of strain SH1-B showed 74.2-87.6% average nucleotide identity (ANI) with the type strains of the closest species. Additionally, digital DNA-DNA hybridization (dDDH) analysis revealed low relatedness, with a maximum value of 34.9% observed for its closest phylogenetic relative, P. khazarica TBZ2(T). The G + C content of the genomic DNA was 62.2%. The major polar lipids were phosphatidylethanolamine (PE), phosphatidylglycerol (PG) and an unidentified phospholipid (PL1). The predominant cellular fatty acids of strain SH1-B included C-18:1 omega 7c, C-16:0,C- C-12:0,C- summed feature 3 (C-16:1 omega 6c/C-16:1 omega 7c). Considering the phenotypic differences and low genomic identity compared to the closest species, strain SH1-B was identified as a novel species within the genus Pseudomonas, for which the name Pseudomonas pichavaramensis sp. nov. is proposed. The type strain is SH1-B-T (= MTCC 13546(T) = JCM 37389(T) = ICMP 25819(T)).
